The organized trafficking of leukocyte subsets through the tissue compartments of the body is critical to the generation of innate and adaptive immune responses and for normal organ immuno-surveillance; however, when misguided, such processes can promote uncontrolled inflammation resulting in tissue pathology. It is now well established that cellular adhesion and chemokine receptors play a central role in regulating tissue-specific leukocyte migration. Recent studies have started to unravel the complex mechanisms underlying the expression of these receptors on lymphocytes and highlight the importance of the local lymph node environments and migratory dendritic cells in the induction of peripheral tissue homing receptors. While immunologists have traditionally tended to focus on primary and secondary lymphoid compartments to study immunological processes and immune cell interactions, there is a growing realization that many, if not most, of our immune cells are located in peripheral tissues and that each of these sites possess unique immunological characteristics. The Keystone Symposia meeting on Chemokines and Leukocyte Trafficking in Homeostasis and Inflammation will bring together scientists and clinicians with expertise in immune cell migration, cellular adhesion/chemokine receptor regulation and tissue-specific leukocyte trafficking with a focus on the peripheral tissue compartments in health and disease.

The meeting is motivated by the hypothesis that these molecules can be developed into effective drugs for the treatment of many common diseases that are collectively responsible for a great deal of human suffering. As this symposium will highlight, there is still much to do, but many in the field now hope that the time has finally arrived when "antisense" therapies will finally come of age and fulfill their promise as the magic bullets of the 21st century.

Hundreds of millions of people world-wide are infected each year with protozoan parasites. The mortality and morbidity associated with these diseases are expected to increase due to the development of drug resistant parasites, the absence of adequate vaccines and a shortage of anti-protozoan agents in the drug development pipelines. Understanding the pathogenesis, mechanisms of growth and differentiation, and metabolic processes of the parasites has provided new insights and challenges in the identification of novel drug targets for malaria, leishmaniasis and trypanosomiasis. Integrated strategies combining target identification and validation with advances in structural biology, genetic manipulations and high-throughput high-content screening platforms are likely to accelerate the development of anti-protozoan drugs. This meeting is the only international meeting that addresses the development of drugs to treat and prevent parasite infections. Parasitologists, chemists, pharmacologists and clinicians from academia, government and industry will come together at this meeting to discuss opportunities for the advancement of novel anti-protozoan chemotypes and progression of lead compounds into clinical development. Much progress has been made in the identification of key signaling pathways regulating blood vessel development. There is also evidence that various stromal cell types participate in normal and pathological angiogenesis in a tissue- or context-specific fashion. However, many important molecular and biological questions regarding these processes remain unanswered. In addition, several angiogenesis inhibitors have been approved by the FDA as therapy for cancer or intraocular neovascular disorders. Perhaps some of the most successful applications of anti-angiogenic therapy are in age-related macular degeneration and other intraocular neovascular diseases, although such findings have been rarely presented outside specialized meetings. The Keystone Symposia meeting on Angiogenesis: Advances in Basic Science and Therapeutic Applications will critically examine the latest basic science on the regulation of angiogenesis in health and disease, discuss the lessons from clinical trials in cancer and intraocular neovascular disorders and identify novel therapeutic targets. Cancer involves the misregulation of cell growth, proliferation, differentiation and death programs. These programs are all regulated at the gene expression level, and chromatin structures and modifications are fundamental regulators of these programs. Indeed, the regulation of many of the most important oncogenes and tumor suppressors operate either largely or in part through a chromatin regulatory mechanism. Furthermore, chromosomal instability and DNA hypomethylation are found in a majority of cancers. Here, one must understand both how these processes normally work and precisely how they are misregulated in cancer in order to design effective and targeted therapies. The 2012 Keystone Symposia meeting on Chromatin Dynamics aims to understand the mechanistic relationships between chromatin and transcription as well as between chromatin and DNA repair and stability, processes at the heart of cancer initiation, prevention, and treatment. Core processes of epigenetic inheritance include DNA methylation, histone modification, nucleosome remodeling, nuclear dynamics and chromatin interaction with non-coding RNAs. Dr. Elaine Larson, Associate Dean for Research and Professor of Pharmaceutical and Therapeutic Research at the Columbia University School of Nursing and Professor of Epidemiology, Columbia University Mailman School of Public Health, has been selected to give the second NINR Director's Lecture. Larson, a member of the National Advisory Council for Nursing Research, will discuss her globally recognized research in infection prevention and control in a lecture titled “Infection Prevention: An Interdisciplinary Team Approach. Dr. Larson is a former Dean, Georgetown University School of Nursing. She has been a member of the Board of Directors, National Foundation for Infectious Diseases, and the Report Review Committee, National Academy of Sciences. She is a fellow in the Institute of Medicine and the Director of the Center for Interdisciplinary Research to Prevent Antimicrobial Resistance at Columbia University and has been Editor of the American Journal of Infection Control since 1995. She has published more than 250 journal articles, four books and a number of book chapters in the areas of infection prevention, epidemiology, and clinical research. The NINR Director's Lecture is an annual event, designed to bring the nation's top nurse scientists to the NIH campus to share their work and interests with a trans-disciplinary audience. The award was initiated as part of the year-long observation of the Institute's first 25 years at the NIH.

MRI-based cell tracking is one of the most exciting and broadly impacting disciplines within radiology. Major advances in nanoparticle technology, coupled with advances in MRI technology, have yielded enhanced detection capabilities with sensitivity of individual cells in vivo. Novel probes and molecular biology provide the opportunity to monitor cell function and gene expression in vivo. Clinically important paradigms, such as stem cell transplantation, inflammation, cancer and regenerative medicine have benefited immensely from established protocols for using MRI-based cell tracking. Furthermore, a wealth of information has been garnered in basic sciences; in neuroscience, immunology and development, owing to MRI-based cell tracking.  Recently, MRI-based cell tracking has graduated from use as a research tool to clinical applications. While there have been only a few studies in humans, this progress is encouraging. This workshop is intended to bring together experts in nanoparticle technology, MRI and molecular and cell biology with biologists, biomedical engineers and physicians in an effort to share information across disciplines and accelerate the translation of MRI-based cell tracking to clinical settings.
